Key stakeholders’ perspectives on the ideal partnering culture in construction projects

摘要: This paper examines the current state of project cultures in the German turnkey construction industry and the ideal project cultures in terms of partnering from the perspective of various key stakeholders (i.e., Investors, General Contractors, (Sub-)Contractors and Designers). To investigate the current and ideal cultures, data were gathered among the key stakeholders by means of a survey study with 72 respondents divided over 12 companies. The respondents rated the current and desired cultures by using the Organizational Culture Assessment Instrument, which belongs to the Competing Values Framework. The investigations show many similarities and differences between the stakeholder perspectives of the current and the idealized partnering project cultures. Mainly, the General Contractors desire more cooperative behaviors than the (Sub-)Contractors, and the Investors desire more pronounced flexibility than the General Contractors. All stakeholders desire a cultural change from highly competitive behaviors toward more cooperation. Changes in terms of clear procedures or more flexibility are only desired by the Designers. Defining both the current and an ideal partnering project culture enables academics and project managers to compare their actual project cultures to an ideal situation. With such an approach, academics and project managers could measure whether new tools or changes in resources affect their project cultures toward a partnering project culture.

Construction engineering management culture shift: Is the lowest tender offer dead?

摘要: The procurement of public construction projects must walk a fine line between the corruption of state officials and collusion of contractors. The method of awarding projects to the lowest responsible tenderer was originally implemented to guard against corruption of state officials. However, an investigation of the construction industry in the Canadian province of Quebec showed that lowest-tender-offer procurement gave rise to collusion of companies tendering for the contracts. Alternatively, best-value procurement has been used for decades, but here problems arise owing to the necessity of subjective judging of measures other than price to compare bids, giving rise to time- and money-consuming protests. The paper proposes a compelling argument that the construction engineering management (CEM) culture should refocus its efforts on enhancing project cost certainty rather than merely searching for means to design a project in a manner that produces the lowest initial cost, and awards the construction to the lowest tender offer that focuses on cost savings during the project development and delivery process. The difference in the two approaches is subtle but extremely important. To make the transition, the engineering management tools must be advanced to the next level. This means that all project control tools for managing cost, schedule, and technical scope must be transformed from working in the deterministic mode to the stochastic mode, thus making the probability of completing the project within or below its official budget the primary decision criterion. To do so, CEMs must accept that there is a benefit in paying more for an alternative that increases cost certainty for the entire project. The authors of this paper hope that it will provide the grist for a more general dialog across all industry sectors where engineering management is practiced.

Decellularized extracellular matrix mediates tissue construction and regeneration

摘要: Contributing to organ formation and tissue regeneration, extracellular matrix (ECM) constituents provide tissue with three-dimensional (3D) structural integrity and cellular-function regulation. Containing the crucial traits of the cellular microenvironment, ECM substitutes mediate cell–matrix interactions to prompt stem-cell proliferation and differentiation for 3D organoid construction in vitro or tissue regeneration in vivo. However, these ECMs are often applied generically and have yet to be extensively developed for specific cell types in 3D cultures. Cultured cells also produce rich ECM, particularly stromal cells. Cellular ECM improves 3D culture development in vitro and tissue remodeling during wound healing after implantation into the host as well. Gaining better insight into ECM derived from either tissue or cells that regulate 3D tissue reconstruction or organ regeneration helps us to select, produce, and implant the most suitable ECM and thus promote 3D organoid culture and tissue remodeling for in vivo regeneration. Overall, the decellularization methodologies and tissue/cell-derived ECM as scaffolds or cellular-growth supplements used in cell propagation and differentiation for 3D tissue culture in vitro are discussed. Moreover, current preclinical applications by which ECM components modulate the wound-healing process are reviewed.

Studies on culture condition of new marine bacterium sp. SY01

摘要: New marine bacterium sp. SY01, producer of prodigiosin, was isolated from the seawaters of Sanya Bay. The culture conditions of this bacterium were investigated. sp. SY01 was cultured in 2216E media which contained tryptophan, histidine, lactonic acid, camphor, limonene, casein, diphenyl guanidine, coumarin and 1,3-dinitrobenzene, respectively. After 5 days cultivation, the extracts of different culture broths were detected by direct infusion mass spectroscopy using positive ESI mode. As the results, tryptophan, histidine and casein didn’t show any observable influences on the biosynthesis of prodigiosin. Lactonic acid, camphor, limonene, diphenyl guanidine, coumarin could inhibit the bacterium growth and prodigiosin biosynthesis to a certain extent, slower the culture broth to turn red. However, 1, 3-dinitrobenzene inhibited the bacteria to produce prodigiosin completely. MS data suggested that various metabolites with chemodiversity were produced in different culture media. In particular, a series of high-molecular-weight compounds with high relative abundances were observed in the medium containing limonene. To further optimize the culture condition, more new prodigiosin analogues and lead compounds can be obtained and the goal of “one strain-many compounds” can be achieved.

Investigation of polyhydroxyalkanoates (PHAs) biosynthesis from mixed culture enriched by valerate-dominant

摘要: Valerate-hydrolysate enriched culture showed great ability to produce 3HV and 3H2MV. Valerate-hydrolysate enriched culture had more in the community. Mixed iso-/n-valerate was effective at balancing microbial growth and PHAs synthesis. Co-substrates of valerate and propionate raised the fraction of 3HV and 3H2MV. The production of polyhydroxyalkanoates (PHAs) with a high fraction of 3-hydroxyvalerate (3HV) and 3-hydroxy-2-methylvalerate (3H2MV) from mixed culture enriched by valerate-dominant hydrolysate was evaluated in this study. After long-term enrichment, the culture showed strong ability to synthesize 3HV and 3H2MV, even with acetate-dominant substrate. The ultilization of single or mixed iso-/n-valerate by the enriched culture showed that the mixture of iso-valerate and n-valerate was more efficient substrate than any single in terms of balancing microbial growth and PHAs synthesis. Besides, through comparing the kinetics and stoichiometry of the tests supplying valerate and propionate, the enriched culture with equivalent valerate and propionate (1:1 molar ratio) exhibited superior PHAs production performances to pure valerate or propionate, attaining more than 70 mol% of 3HV and 3H2MV. The above findings reveal that valerate-dominant hydrolysate is a kind of suitable substrate to enrich PHAs producing culture with great capability to synthesize 3HV and 3H2MV monomers, thus improving product properties than pure poly(3-hydroxybutyrate) (P3HB); also 3HV and 3H2MV production behaviors can be regulated by the type of odd-carbon VFAs in the substrate.

已有大量研究对施工中的返工问题进行了研究,但解决返工问题的成效有限,仍然困扰着施工进程,这对项目的绩效产生不利影响。几乎所有返工研究都侧重于确定其近端或根本原因,因此忽略了导致返工的条件。本文借鉴了之前的实证研究,对这一空白进行了研究,以提供一个急需的理论框架,从而更好地理解返工原因、返工后果以及如何在施工期间减少返工。从审查中得出的理论框架使施工组织认识到,减少返工应首先建立一套差错管理文化,包括真正的领导力、心理安全、差错管理导向和复原力。本研究认为,一旦在施工组织及其项目中建立了差错管理文化,施工组织就能更好地认识用于解决返工问题的方法、工具和技术的益处,如精益生产®和建筑信息模型。本文还为未来研究提供了方向,并确定了对实践的意义,以便能够在施工中减少返工。

Positive stool culture could predict the clinical outcomes of haploidentical hematopoietic stem cell

摘要: We aimed to identify the effect of positive stool cultures (PSCs) on the clinical outcomes of patients undergoing haploidentical hematopoietic stem cell transplantation (haplo-HSCT) ( = 332). PSCs were observed in 61 patients (PSC group, 18.4%). Enterobacteriaceae in stool specimens was associated with a higher risk of bloodstream infection, and in stool specimens was related to a higher risk of platelet engraftment failure. The cumulative incidence of infection-related mortality 1 year after haplo-HSCT in the PSC group was higher than that of the patients who showed persistently negative stool cultures (NSC group; 19.2% vs. 8.9%, = 0.017). The probabilities of overall survival (71.4% vs. 83.8%, = 0.031) and disease-free survival (69.6% vs. 81.0%, = 0.048) 1 year after haplo-HSCT for the PSC group were significantly lower than those for the NSC group, particularly for patients who had in their stool specimens. In multivariate analysis, in stool specimens significantly increased the risk of mortality and was associated with poorer survival. Our results showed that PSC influenced the clinical outcomes after haplo-HSCT, particularly those who had in their stool specimens.

摘要: The role of on the central carbon metabolism was investigated through analyzing the deficiency of this regulon gene under aerobic and glucose-enriched culture conditions. The experimental results showed that while the wild type cells exhibited an overflow metabolism effect, the -deleting mutation alleviated this effect with the significant suppression of acetate accumulation under a high glucose condition. This gene deletion also induced the twofold upregulation of and one-tenth downregulation of , respectively. The overflow metabolism effect was confirmed to be recovered by re-introducing gene into the mutant. These results demonstrated changed the central carbon metabolism toward acetate overflow through dehydrogenation of pyruvate and reduction of TCA cycle activity.

Comparison of human nasal epithelial cells grown as explant outgrowth cultures or dissociated tissue cultures

The purpose of this study was to compare cell growth characteristics, ciliated cell differentiation, and function of human nasal epithelial cells established as explant outgrowth cultures or dissociated tissue cultures. Human nasal mucosa of the uncinate process was obtained by endoscopy and epithelial cell cultures were established by explant outgrowth or dissociated tissue culture methods. Epithelial cell growth characteristics were observed by inverted phase contrast microscopy. Ciliated cell differentiation was detected by β-tubulin IV and ZO-1 immunocytochemistry. Basal and ATP-stimulated ciliary beat frequency (CBF) was measured using a high-speed digital microscopic imaging system. Both the explant and dissociated tissue cultures established as monolayers with tight junctions and differentiated cell composition, with both types of cultures comprising ciliated and non-ciliated epithelial cells. Fibroblasts were also frequently found in explant cultures but rarely seen in dissociated tissue cultures. In both culture systems, the highest ciliated cell density appeared at 7th–10th culture day and declined with time, with the lifespan of ciliated cells ranging from 14 to 21 days. Overall, 10% of the cells in explant cultures and 20% of the cells in the dissociated tissue cultures were ciliated. These two cultures demonstrated similar ciliary beat frequency values at baseline (7.78±1.99 Hz and 7.91±2.52 Hz, respectively) and reacted equivalently following stimulation with 100 μM ATP. The results of this study indicate that both the explant outgrowth and dissociated tissue culture techniques are suitable for growing well-differentiated nasal ciliated and non-ciliated cells, which have growth characteristics and ciliary activity similar to those of nasal epithelial cells in vivo.

摘要: This work is aimed to achieve the optimal hyaluronic acid (HA) production by batch culture of via the supplement of nucleotide bases using response surface methodology (RSM). First, the influence of nucleotide bases (adenine, guanine, cytosine, thymine, and uracil) on microbial HA production was investigated using fractional factorial design (FFD). By a 2 FFD, uracil was found to be the most significant factor for cell growth and HA production, while the other nucleotide bases were shown to have no significant effects on cell growth and HA production. Also, the impact of uracil on cell growth and HA production was further investigated by RSM, where two variables were considered: uracil concentration and supplement time. The optimal uracil concentration and supplement time were found to be 0.051g/L and 7h, respectively, and the predicted maximal HA production reached 6.42g/L. The maximal HA production increased from 5.0g/L of the control without uracil supplement to 6.31g/L at the optimal conditions in validation experiments.
